Cover: The cover image shows ribbon representations of selected parts of the flavoprotein medium-chain acyl-CoA dehydrogenase (MCAD). The cofactor FAD and the substrate analog (3-thiaoctanoyl-CoA) are depicted in yellow and purple, respectively. Side-chain interactions in the local environment of three amino acid residues affected by missense mutations were analyzed to delineate their functional and conformational impact. A mutation at position 133 (Y133H) affects the structural integrity of the active site, whereas R181C disturbs the interconnection of the b-domain with the C-terminal a-domain of the protein, and R388S alters the conformation of the active site entrance (clockwise direction from top).
